A plasmid 3 Kb in length contains a gene for ampicillin resistance and a gene for tetracycline resistance. The plasmid has a single site for each of EcoRI, Bg/II, and PstI. If DNA is cloned into the EcoRI site, resistance to either antibiotic is not affected. DNA cloned into the PstI site eliminates ampicillin resistance.DNA cloned into the Bg/II site abolishes terracycline resistance. Complete double digestion fragment produced are as follows: EcoRI + PstI=2.3 and 0.7 Kb; EcoRI + Bg/II=2.77 and 0.3 kb.
